Interaction of liposomes on endothelial cells
نویسنده
چکیده
Liposomes are small vesicles (∼100 nm) composed of various of lipid molecules which build their membrane bilayers. Different forces acting on and among lipid molecules in liposome membrane influence their dynamics and interactions with other particles (e.g. cells). In the case of liposome interacting with endothelial cells (e.g. blood-brain barrier) various of transport mechanisms have to be considered (e.g. receptor-mediated transcytosis). Liposome-cell interaction which is exchange of lipids, absorbtion of liposomes on cell membranes and specially fusion or endocytosis, can be described with proper equations of mass action. Furthermore, experimental measurements of liposome-cell interaction on MDCK cells with similar charachteristics compare to actual endothelial cells in BBB were made. The measurements showed the correlation of specific membrane properties with the rate of interaction or transcytosis, which could lead to an optimal liposomal conformation to effectively traverse BBB.

Background: The innate and adaptive immune responses are dependent on the migration of leukocytes across endothelial cells. Dendritic cells (DCs) play an important role in the initiation of cellular immune responses during their migration from tissues into the lymph nodes where they interact with endothelial cells of lymphatic vessels. We investigated the effect...

Objective(s): Dendritic cells (DCs) play a critical role in activation of T cell responses. Induction of type1 T helper (Th1) immune response is essential to generate protective immunity against cutaneous leishmaniasis. The intrinsic tendency of liposomes to have interaction with antigen-presenting cells is the main rationale to utilize liposomes as antigen carriers. In the present study, the e...

BACKGROUND/AIMS In designing a versatile liposomal drug carrier to hepatic stellate cells (HSC), the interaction of mannose 6-phosphate human serum albumin (M6P-HSA) liposomes with cultured cells was studied. METHODS M6P-HSA was covalently coupled to the liposomal surface and the uptake and binding of 3H-labelled M6P-HSA liposomes by primary rat HSC and liver endothelial cells was determined....

Biological processes such as endocytosis/exocytosis, interactions between cells and the dynamic reorganization of the cytosqueleton are between the topics extensively studied using the Atomic Force Microscopy (AFM) technique. The topographical structure of the cell membrane itself is complex, rich in natural corrugations and ridges. These surface characteristics obscure the identification of ex...
متن کاملSynthesis and evaluation of stearylated hyaluronic acid for the active delivery of liposomes to liver endothelial cells.
Hyaluronic acid (HA) is a naturally-occurring ligand that can be useful for targeting liver endothelial cells. We describe herein the development of a new HA-lipid conjugate for the efficient delivery of liposomes to liver endothelial cells. When free HA coated cationic liposomes were injected into mice, their accumulation in the liver was significantly decreased depending on the content of fre...
